Significance The functional diversity of MAPKs is dependent on differential selection substrates for phosphorylation. Nevertheless, the mechanisms assisting to maintain substrate specificity in plants are not known. Our study shows that WRKY33, a pathogen-responsive MPK3/6 substrate, SUMOylated response pathogen-associated molecular patterns and pathogen signals. SUMOylation mediates WRKY33 phosphorylation its activation. This correlation reliant selective identification by via SUMO interaction motif (SIM) sites. Therefore, important selectively interact with SUMO–SIM module undergo thus data reveal mechanism which prioritize pathways differentially selecting using module.
